Oxytocin linked to language growth in young autistic children, study finds

A new study reveals that higher levels of oxytocin in young autistic children are associated with improved language and communication skills, highlighting potential therapeutic avenues.

A study published in Frontiers in Psychiatry has found that higher oxytocin levels in young autistic children were linked with stronger language and communication skills, while vasopressin showed no clear relationship. The analysis drew on data from 148 children aged 3 to 7.5 years and suggests that the hormone may be more closely tied to vocabulary growth and expressive communication than previously understood.

The research team examined blood samples from the larger group and saliva samples from a second cohort of 40 children aged 2 to 6 who had taken part in a six-week trial of intranasal oxytocin followed by positive social interaction. In the first cohort, baseline plasma oxytocin was associated with performance on vocabulary tests and with verbal and non-verbal communication ratings, even after accounting for age and sex. The link was stronger for spoken language than for comprehension. By contrast, vasopressin was not significantly related to any of the language measures.

A smaller analysis that added broader developmental scores as a control found the same pattern, though some associations weakened. The authors also explored possible sex differences. Boys showed clearer links between oxytocin and language measures, while the much smaller female sample hinted at different patterns, including a stronger connection between oxytocin and non-verbal communication. The researchers said those findings should be treated cautiously because the number of girls was limited.

In the intervention cohort, children who received oxytocin showed better scores on several language-related scales than they did on placebo, and gains were associated with rises in saliva oxytocin after treatment. Two of the measures were also indirectly mediated by those hormone changes, suggesting the treatment effect may have run partly through increased oxytocin levels. The authors said the findings add to evidence that oxytocin could have therapeutic potential for autistic children with language difficulties, although they stressed that larger studies are needed.
